Abstract

Obesity induced by a high-fat diet (HFD) is fundamentally linked to chronic systemic inflammation, which is clinically characterized by elevated levels of C-reactive protein (CRP). Objective: This research evaluated the anti-inflammatory efficacy of Nigella sativa (black seed) oil in HFD-fed Wistar rats (n=25). the 25 male Wistar rats randomly assigned into 5 groups: normal, negative control, positive control, treatment 1 (regular dose of black seed oil) and treatment 2 (High dose black seed oil). The result confirmed that the HFD control group exhibited a significant surge in CRP concentrations (2.28 ± 0.12 ng/mL) relative to the normal control group (0.74 ± 0.03 ng/mL, p < 0.001). Specifically, the high-dose intervention (108 mg/kg body weight) demonstrated superior efficacy over the low-dose regimen (54 mg/kg), substantially reducing CRP to 0.85 ± 0.02 ng/mL. This equates to a 63% reduction compared to the HFD control (p < 0.001). Statistical validity across these metrics was confirmed via a one-way ANOVA with a Tukey post-hoc test (F = 145.2, p < 0.001). 108 mg/kg as the optimal dose for CRP suppression, highlighting the robust therapeutic viability of Nigella sativa in managing obesity-related inflammation.
