Validation of Patchouli Oil Alcohol Content Analysis Method Using Gas Chromatography-Tandem Mass Spectrometry (GC-MS/MS)

Abstract

Validation of the analytical method is an important step to ensure the method produces accurate, precise, and reliable data. This study aims to validate the method for determining patchouli alcohol (PA) levels in patchouli oil using GC-MS/MS and apply it to samples. The analysis was carried out using Multiple Reaction Monitoring (MRM) mode, monitoring the ion m/z 123 as a quantifier and m/z 109 as a qualifier. PA standard solutions were prepared in the range of 2.5–20.00 ppm in n-hexane, with a retention time of 8.60 minutes and an analysis time of approximately 11.3 minutes. The validation results showed good method performance, characterized by high linearity (R² ≥ 0.9984), accuracy (% recovery 98.00–101.89%), and qualified precision (%RSD ≤ 1.212% for intraday and ≤ 1.663% for interday). The LOD and LOQ values were 0.74 ppm and 2.46 ppm, respectively. The validated method was then applied to patchouli oil samples from three locations: North Kolaka (44.72%), Kolaka (42.85%), and East Kolaka (32.90%), and showed PA levels that met the Indonesian National Standard (SNI) quality standard of >30%. Thus, the GC-MS/MS method was declared valid and suitable for use in analyzing patchouli alcohol levels in patchouli oil.
